Can I add a NTFS drive as a second drive?

I have tried searching the forum for the answer to this question but have been unable to find a simple clarification.
I am planning to add a second drive to my HDVR2 using the TwinBreeze kit I have bought. The drive I am adding is currently formated as a NTFS drive in my PC.
Can I just add it using MFS tools or do I have to reformat it as a FAT32 drive first?
Thanks for your help!

You can just use MFSTools. It does everything for you.

The tools will overwrite whatever might have been on the drive. So it really does not matter if it is new or used, once MFSTools starts it thing, it wipes the drive, so nothing to worry about.
